Organelle Reuse by Macrophages During Dead Cell Clearance

Abstract Billions of cells die every day in our bodies which are promptly and safely disposed of by macrophages which are sentinel cells of the immune system by a specialised process called "efferocytosis". This fundamental process is critical for maintaining tissue homeostasis during health and disease. Following efferocytosis, macrophages must deal with the enormous load of the newly acquired dead cell-associated biological material which include lipids, proteins, DNA, and cellular organelles. The current view is that all these materials are degraded within the macrophage and the constituent building blocks are recycled for use by macrophages or exported through dedicated efflux or transport channels for disposal. Generation of new cellular organelles is an energy intensive process. Since cell-to-cell transfer of organelles is an established phenomenon, we postulate that macrophages could potentially acquire organelles from dead cells and reuse them. In this proposal, we will challenge the prevailing view that all dead cell-associated biological materials are degraded by testing our novel hypothesis that certain cellular organelles may escape this fate and instead be reused by macrophages for cellular function. These findings will open fundamentally new areas of research in phagocyte biology and organelle reuse in physiology.
